Mrs Johnson said that the thieves entered the building through the temporary security doors that were installed after the previous robbery in August of last year.
Mrs Johnson said that whoever it was seemed to know what they were doing.
"They cut the power and broke through the door in the early hours of Monday morning," she said.
"It's a bit different to last time in that they seemed to know what they were looking for and where to find it."
The target of the robbers were cigarettes, cash and electronic equipment.
Over $20,000 worth of cigarettes were stolen.
Mrs Johnson said that until she's finalised a stock-take the total amount stolen or damaged was unknown.
"There were lollies strewn across the floor and drinks taken from the Coke fridge and the Dairy Farmers fridge," she said.
This recent break-in comes after a spate of crimes across the shire including theft of $800 from a vehicle in Charles Street Blayney, a robbery at the Forest Reefs Tavern and four business break-ins in Blayney.
Sergeant Karl Hutchings said that investigations into the Millthorpe Corner Store break-in are continuing.
"We're asking the community, and in particular any of the Cadia mine workers who travel through Millthorpe, that if they saw anything suspicious on Monday morning to call us at Blayney police on 6368 9599 or Crime Stoppers on 1800 333 000."
